Het seizoen 1965/1966 van de Nederlandse Eredivisie was het tiende seizoen waarin onder de KNVB-vlag een professionele Eredivisie werd gevoetbald. Dit seizoen streden zestien clubs in de Eredivisie. Nieuw door promotie uit de Eerste divisie waren Willem II en Elinkwijk. Zij namen de plaats in van NAC en Sittardia, die gedegradeerd waren.

Daarnaast was dit het eerste seizoen voor de fusieclub FC Twente '65, waarin SC Enschede en Enschedese Boys waren opgegaan. De ploeg kreeg de Eredivisieplaats van SC Enschede.

Omdat het volgende seizoen weer met achttien clubs zou worden gespeeld, degradeerde er in 1965/66 één club. Ajax, dat het vorig seizoen zijn slechtste prestatie ooit in de Eredivie had gehaald, werd landskampioen. Heracles degradeerde rechtstreeks.
