De club werd in 1926 opgericht en promoveerde in 1975 voor het eerst naar het hoogste niveau. Na enkele jaren in de lagere divisies promoveerde de club in 2009 naar de Beta Ethniki en werd daar in 2011 kampioen. Hierdoor speelt Pantetolikos in het seizoen 2011/12 andermaal in de Alpha Ethniki (Super League). Al na één seizoen volgde opnieuw degradatie naar de Beta Ethniki maar weer eens seizoen later keerde de club weer terug op het hoogste niveau.
